Phase 1 Study of SYNC-T Therapy SV-102 Reveals Promising Survival Rates in Prostate Cancer
KEY TAKEAWAYS
1

Median overall survival not reached in the overall patient population after 29.6 months of follow-up.

2

80% overall survival rate at 24 months for all patients, with 100% in the hormone-sensitive cohort.

The investigational therapy was well tolerated, with no severe immune-related adverse events reported.

A recent phase 1 clinical trial has shown promising survival outcomes for SYNC-T Therapy SV-102, an innovative in situ immunotherapy for metastatic prostate cancer. The study, led by Dr. Carlos A. Vargas, was presented at the European Society for Medical Oncology Congress 2026, highlighting its potential in improving patient survival rates.

SYNC-T Therapy SV-102 employs a unique mechanism combining partial tumor cryolysis with the delivery of immunomodulators, including anti–PD-1 and anti–CTLA-4 monoclonal antibodies. This approach aims to stimulate a systemic immune response by releasing tumor-specific antigens while preserving the tumor structure. The study enrolled 15 patients, with a mix of those suffering from metastatic castration-resistant prostate cancer (mCRPC) and metastatic hormone-sensitive prostate cancer (mHSPC).

In the updated analysis, the median follow-up period was 29.6 months, during which the overall survival (OS) rate for all participants reached 80% at 24 months. Specifically, patients in the mCRPC group had a median OS of 27.9 months, while the mHSPC cohort reported a 100% OS rate at the same interval. Notably, three out of six patients who achieved a complete response remained disease-free for over 26 months.

The safety profile of SYNC-T Therapy SV-102 was also encouraging, with 95% of treatment-related adverse events classified as grade 1 or 2. No severe immune-related adverse events were documented, indicating a favorable tolerability for patients undergoing this treatment. These findings have prompted further investigation in the ongoing multicenter phase 2 LEGION-100 trial.

Looking ahead, the promising results from this phase 1 study may pave the way for more extensive trials and potential approval of SYNC-T Therapy SV-102 as a viable treatment option for metastatic prostate cancer. The medical community is keenly observing the outcomes of the upcoming phase 2 trial, which could significantly impact treatment protocols for prostate cancer patients worldwide.
